Abstract

Chromatography is simply a method of separation of a mixture in to individual components by using stationary and mobile phase. Tswett and Days experiments on colored plant pigments described in 1903 – 1906 are the backbone of chromatographic techniques. The ion exchange, surface adsorption, partition, and size exclusion chromatographic techniques are based on molecular characteristics and interaction. The simple chromatography techniques like column, thin layer, and paper chromatography are based on the stationary bed. Since ancient time herbs and herbal formulations are used for their medicinal usage as they contain various types of bioactive compounds. Extraction, isolation and characterization of active chemical constituents in botanicals and herbal preparations is possible only with the help of chromatography. The analysis of bioactive chemical constituents involves chromatographic techniques such as column chromatography, HPLC and TLC etc. Chromatographic techniques are useful in analysis and separation of biological samples. They have enhanced the development of new procedures in the field of pharmaceutical analysis. The newly developed methods, allow for more accurate testing of raw materials, intermediates and final products. The chromatographic techniques also enable quality control of pharmaceuticals and herbal formulations. Chemical constituents isolated from medicinal plants, provided an opportunities for new drug leads for further development of drugs.
